Cloning and Nucleotide Sequencing of Porcine Encephalomyocarditis Virus

Porcine encephalomyocarditis virus(EMCV) causes a fatal disease in young pigs. Recently it has been known as a cause of a swine reproductive failure and virus was isolated from fetal samples from swine herds with a history of reproductive
problems
in
Korea. One cDNA clone prepared from EMCV Korean isolate was identified and its nucleotide sequence was determined. This clone was located in the 3 region of the gene. Its nucleotide sequence and deduced amino acid sequence were compared with
published
sequences of the Palmenberg strain and D-strain It had 99.2% and 89.5% nucleotide sequence homology with the Palmenberg strain and Dstrain, respectively. A probe prepared from this clone was EMCV -specific and used in a slot blot hybridization a
in
situ
hybridization assay to detect EMCV.
